I remember you said that there are some VMs from the same problem datastore and its working fine rite? If other VMs are working fine and even after vmotion if it is not working then this should be something wrong with the flat vmdk.
Lets try one more thing here. Power on the problem VM from different host, take note of the error time stamp then post the vmkernal/messages logs which cover that time.
